The sample is very complex and contains a lot of impurities that are difficult to separate. In order to increase output while ensuring product purity, a two-step preparation method is used. First, a large-diameter preparative column is used to roughly divide the sample to achieve separation of the target from most impurities. Then take the coarsely divided product as a sample, use a small-diameter preparative column, lengthen the column length of the chromatographic column in series to improve the column efficiency, and perform the second preparative separation.
